If by over you mean the Lakers becoming the laughing stock of the franchise, it would have to be signing Md'T over KFC. In retrospect, KFC wouldn't have made a difference as his antiquated system wouldn't have led the Lakers to success anyways, but that move showed that ownership was making decisions on a personal perspective over a business perspective.
However, if you mean over as in the Lakers not going to ring for a while, it would have to be Kobe and KFC publicly humiliating MVPau after the Mavs series. You can't have a championship team where the alpha gets all the blame and non of the glory, especially one as emotionally fragile as MVPau, who would one day just say "screw this, I have sacrificed enough to prop up the beta, let him deal with this". And that was exactly what happened.
