Not going to argue on that it just going to be really hard to keep up with new variants at some point shit could turn South real quick - Lock downs, Civil Unrest, etc. It doesn’t take much to move a civilized society to basic barbaric real quick - I am not talking about people against vaccine more of economy shutting down - People loosing jobs, Inflation out of control, supply side issues getting basic food and medicine to people. I would hate to be living in a major city during these times.

Sound like the plot for a cheezy Bollywood movie, tbh. Odds of that happening after vaccines are available (and the upcoming treatments like the pills) are fairly low.

It actually does take quite a bit for an actual civilized society to turn barbaric. I mean, this isn't even close to the first time we have to deal with a pandemic like this one and none of those things happened. Not now, not 100 years ago with the Spanish Flu.

The economic malaise is real though, but that's why it's important to have an active government in times of emergencies.

I would actually rather be living in a major city than the backwoods, close to the best hospitals and clinics with the latest equipment and treatments, tbh... but that's just my preference.

He's lucky his horrific response to the pandemic distracted away from his attempt to get us into the decades long neocon wet dream war with Iran. And we're lucky Iran acted like the adults in the room.

Adults who shoot down their own airliners.

This is what centers for disease control and the WHO are for. There are plenty tracked viruses we don't hear about because we got them under control. Right now it's the news media looking to pimp fear. You have some here who just love it when there's new fear porn to regurgitate and warn about. Everyone's going to die eventually.

30% is now considered effective? Monoclonals as of today are around ~80% effective, by comparison.

Among more than 1,400 adults in a company study, molnupiravir reduced the combined risk of hospitalization and death by 30%, less than the 50% initially reported based on incomplete results.

Molnupiravir is mutagenic. What could go wrong?

(Merck says rodent studies disclose no problems here.)

Merck’s drug uses a novel approach to fight COVID-19: it inserts tiny mutations into the coronavirus’ genetic code to stop the virus from reproducing.
But that genetic effect has raised concerns that in rare cases the drug could cause birth defects or even spur more virulent strains of the virus.

Damn those are some English size hedges... you got your own vegetative safety system.

I will say SA about 3 weeks ago connected the Salado Creek bike trail to the Leon creek trail. So from one end to the other Im thinking over 40 miles. Down and back, 80 miles. Not like Im going to do this, but pretty awesome for San Antonio. The tough thing is the time change because you have to be off the trails at sundown. I may rework a schedule to get some day riding that is not weekend. Flexibility in a job is something extraordinarily valuable that people should feel lucky to have.

YERUSHALAYIM - Initial data on the Omicron variant received from South Africa and Europe indicate that the current vaccines will continue to be effective against it, according to Israel’s Channel 12 on Tuesday evening. Those who received three doses (including a booster) of anti-coronavirus vaccine can expect 90 percent protection against Omicron, only slightly less than the 90 percent offered against the Delta variant which preceded it, the report said. In addition, it appears that immunity against re-infection by recovering patients is double that against Delta. On the other hand, the first findings do confirm that Omicron may be more dangerous than previous strains for the unvaccinated—2.4 times likely to lead to serious illness than previous strains.

Yep, I ain't ever recovered from my yo-yo weight system. I got down to 159 pound from 245. It was in the era of AIDs so everybody thought I was sick with it. I have pictures where my head didn't lose weight, but my body did. My head looked like it was settin' on a golf tee.

My body has never been the same. I still have the designer jeans collection I bought (size 32 waist) from the Phoenix metro area upscale thrift shops. I can't even get one leg in there now. Nary a one.

Soda crackers and beets. The weight just fell off. It was incredible. I'd lose 5 lbs over night.

There's tons of medicines with side effects, Darrin. Obviously, this is for people that can't get a much safer vaccine due to some other medical reason and are ok with the side-effects.

There's also another competing pill that works in a completely different manner, so we'll see what the side effects for that one are.

I've read both the pills have been extremely effective at preventing death from COVID on trials vs placebo, even when used late in the infection, so that's positive news.

Yeah, that's the other one. Haven't read about Merck's new testing though, thanks for sharing. Obviously it makes it less useful, but there might be a use case for it. They're all tools in a toolbox.

The FDA issued an emergency use authorization (https://www.fda.gov/news-events/press-announcements/coronavirus-covid-19-update-fda-authorizes-new-long-acting-monoclonal-antibodies-pre-exposure) Wednesday for AstraZeneca’s antibody cocktail, Evusheld, for what is known as pre-exposure prophylaxis, or PrEP, against Covid-19.

To date, such laboratory-produced antibodies have been authorized only as early treatment of Covid-19 or as preventive therapy for high-risk people immediately after close contact with someone who has tested positive.

Evusheld can be used as PrEP by people ages 12 and older who are moderately to severely immunocompromised and may not get adequate immune responses from a Covid vaccine (https://www.nbcnews.com/health/health-news/immunocompromised-people-can-receive-4th-covid-shot-cdc-says-rcna3933). The therapy is also an option for the rare people who have histories of severe adverse reactions to a Covid vaccine or its components.

The AstraZeneca therapy involves getting preventive injections as often as every six months. According to a large placebo-controlled clinical trial, the cocktail is about 83 percent effective at preventing symptomatic disease during such an interval.

The Washington Examiner article is essentially a Pfizer press release, so throw a big handful of salt over your shoulder.

Efficacy figures seem to presume prompt access to medical care and quick diagnosis, two big ifs in the USA, given our weak public health system and our abject failure to scale up rapid testing. Availability and cost may be two other impediments.

Still, could be a game changer if the FDA approves and we can get the pills to people fast enough.

COVID-19 vaccine added to list of mandatory vaccines for school in LA

Gov. John Bel Edwards is moving forward with adding COVID-19 vaccines to the required list of school shots, overturning a vote from a legislative panel last week to nix the mandate recommended by the Louisiana Department of Health.

Louisiana and California are the only two states to mandate the vaccine for school children, according to testimony in Louisiana Health and Welfare Committee hearing.

Though Edwards' action will add COVID vaccines to the required list beginning next school year, parents can opt out by signing and submitting a form to their schools as they can for other required immunizations like polio and measles.

The Biden administration vax-only COVID strategy (deemphasizing masks, social distancing, travel restrictions, quarantine, rapid testing, contact tracing, air quality, ventilation, etc.,) was political, so it makes sense the approval process has been too.

For better and for worse, public health policy is politics. Judging by the fruits, the inordinate emphasis on vaccines has not been successful so far at the population level.

Switching is fine safety wise. Although they all target the same wild type spike there may be some variation between the vaccines which theoretically might give you a somewhat broader antibody response. I'm not aware of any actual science on that so it's just a theoretical. You could also consider getting a J&J which is a completely different platform. The only problem with J&J is it's dead in the water in this country so you will be in the dark as far as future studies go. Which is the reason I switched to Moderna for my 2nd dose.

Listening to TWiV, they think ideally you would wait 9 months for your 3rd dose but a minimum 6 months. Since you have had a severe covid infection and are double vaccinated you probably don't need a booster at all. That said, there are no known safety concerns and since you are 65+ (I think) I'd say go for it. FWIW, my own doctor got the moderna booster after originally getting the 2 pfizer doses.

I just clicked through that video but it looked like he was just going off the levels of circulating antibodies induced after each booster. The problem with that approach is there is no correlate of protection for antibody levels so higher doesn't necessarily mean better. Add to that the fact that circulating antibody levels will always decrease after the initial spike following vaccination and I don't think it means a lot. What we really need are studies on the diversity of antibodies generated, memory B cell formation, and memory T cell formation following the booster doses. We'll get them eventually, but that work takes time and $$$$.

SpFN

A series of recently published preclinical study results show that the Spike Ferritin Nanoparticle (SpFN) COVID-19 vaccine developed by researchers at the Walter Reed Army Institute of Research (WRAIR) not only elicits a potent immune response but may also provide broad protection against SARS-CoV-2 variants of concern as well as other coronaviruses.

Scientists in WRAIR’s Emerging Infectious Diseases Branch (EIDB) developed the SpFN nanoparticle vaccine, based on a ferritin platform, as part of a forward-thinking “pan-SARS” strategy that aims to address the current pandemic and acts as a first line of defense against variants of concern and similar viruses that could emerge in the future.

“The accelerating emergence of human coronaviruses throughout the past two decades and the rise of SARS-CoV-2 variants, including most recently Omicron, underscore the continued need for next-generation preemptive vaccines that confer broad protection against coronavirus diseases,” said Dr. Kayvon Modjarrad, Director of the Emerging Infectious Diseases Branch at WRAIR, co-inventor of the vaccine and the U.S. Army lead for SpFN. “Our strategy has been to develop a ‘pan-coronavirus’ vaccine technology that could potentially offer safe, effective and durable protection against multiple coronavirus strains and species.”

Pre-clinical studies published today in Science Translational Medicine indicate that the SpFN vaccine protects non-human primates from disease caused by the original strain of SARS-CoV-2 and induces highly-potent and broadly-neutralizing antibody responses against major SARS-CoV-2 variants of concern including the SARS-CoV-1 virus that emerged in 2002.

SpFN entered Phase 1 human trials in April 2021. Early analyses, expected to conclude this month, will provide insights into whether SpFN’s potency and breadth, as demonstrated in preclinical trials, will carry over into humans. The data will also allow researchers to compare SpFN’s immune profile to that of other COVID-19 vaccines already authorized for emergency use.

“This vaccine stands out in the COVID-19 vaccine landscape,” Modjarrad said. “The repetitive and ordered display of the coronavirus spike protein on a multi-faced nanoparticle may stimulate immunity in such a way as to translate into significantly broader protection.”https://www.dvidshub.net/news/411320/series-preclinical-studies-supports-armys-pan-coronavirus-vaccine-development-strategy

Experts warn a new COVID pill could backfire and 'create breeding ground for mutant viruses'
the potential drawbacks of molnupiravir, including the frightening possibility that it could—in the words of one researcher—"create a breeding ground for mutant viruses," thereby prolonging the pandemic and adding to its grisly death toll.

"The problem with molnupiravir lies in its mechanism of action,"

"Unlike any previous antiviral drug, molnupiravir does only one thing: It introduces mutations into the viral genome."

molnupiravir relies on inducing even more mutations so that eventually the virus' proteins are damaged beyond function.

while molnupiravir works to some extent, it has not worked very well in Covid-19 patients,"

Merck's pill has proven far less effective in preventing hospitalization than Pfizer's (https://www.commondreams.org/news/2021/12/22/fda-authorization-pfizers-covid-pill-leads-fears-global-medical-apartheid-part-two),

"As long as molnupiravir is in use somewhere in the world,

it could generate repeated cycles of new variants,

with people desperately taking the drug to fight the new variants it spawns,

creating a vicious positive feedback loop while causing more suffering and deaths...

The FDA and Merck have essentially engaged the public in a gamble without public debate."

Congrats to Texas Children's Hospital, the Center for Vaccine Development, and the National School for Tropical Medicine at Baylor College of Medicine.

A cheap scalable vaccine that doesn't require an elaborate cold chain is still needed to tamp down disease and help prevent the emergence of new variants.

HOUSTON, TX (December 28, 2021) – Texas Children’s Hospital (https://www.texaschildrens.org/) and Baylor College of Medicine announced today that CORBEVAX™, a protein sub-unit COVID-19 Vaccine, whose technology was created and engineered at its Center for Vaccine Development (https://www.bcm.edu/departments/pediatrics/divisions-and-centers/tropical-medicine/research/vaccine-development) (CVD), has received Emergency Use Authorization (EUA) approval from the Drugs Controller General of India (DCGI) to launch in India with other underserved countries to follow.

Dubbed “The World’s COVID-19 Vaccine”, it uses a traditional recombinant protein-based technology that will enable its production at large scales making it widely accessible to inoculate the global population. The initial construct and production process of the vaccine antigen was developed at Texas Children’s Hospital CVD, led by co-directors Drs. Maria Elena Bottazzi and Peter Hotez and in-licensed from BCM Ventures (https://www.bcm.edu/about-us/bcm-ventures), Baylor College of Medicine’s integrated commercialization team, to Hyderabad-based vaccine and pharmaceutical company Biological E. Limited (https://biologicale.com/) (BE).

CORBEVAX™ after completing two Phase III clinical trials involving more than 3000 subjects was found to be safe, well tolerated and immunogenic:

CORBEVAX™ demonstrated superior immune response in comparison with COVISHIELDTM vaccine when assessed for Neutralizing Antibody (nAb) Geometric Mean Titers (GMT) against the Ancestral-Wuhan strain and the globally dominant Delta variant. CORBEVAX™vaccination also generated significant Th1 skewed cellular immune response.

CORBEVAX™ nAb GMT against Ancestral-Wuhan strain is indicative of vaccine effectiveness of >90% for prevention of symptomatic infections based on the Correlates of Protection assessment performed during Moderna and Astra-Zeneca vaccine Phase III studies.

CORBEVAX™ nAb GMT against the Delta strain indicates a vaccine effectiveness of >80 percent for the prevention of symptomatic infections based on published studies.

While none of the subjects who took CORBEVAX™ or COVISHIELD™ had serious adverse events, CORBEVAX™ had 50 percent fewer adverse events than COVISHIELDT™.

In the continuous monitoring of phase II studies, CORBEVAX™ showed high persistence of immune response as indicated by <30% drop in nAb GMT till 6 months second dose as compared to >80% drop observed with majority of the vaccines.

“Protein-based vaccines have been widely used to prevent many other diseases, have proven safety records, and use economies of scale to achieve low-cost scalability across the world,” said Dr. Maria Elena Bottazzi (https://www.bcm.edu/people-search/maria-bottazzi-18431), Professor and Associate Dean of the National School of Tropical Medicine (https://www.bcm.edu/education/national-school-of-tropical-medicine) at Baylor and Co-Director of the Texas Children’s Hospital Center for Vaccine Development. “Our decade-long studies advancing coronavirus vaccine prototypes has led to the creation of this vaccine, which will fill the access gap created by the more expensive, newer vaccine technologies and that today are still not able to be quickly scaled for global production.”

The need for safe, streamlined, low-cost vaccines for middle- to low-income countries is central to the world’s fight against the COVID-19 pandemic. Without widespread vaccination of populations in the Global South, additional virus variants will arise, hindering the progress achieved by currently available vaccines in the United States and other Western countries.

“This announcement is an important first step in vaccinating the world and halting the pandemic. Our vaccine technology offers a path to address an unfolding humanitarian crisis, namely the vulnerability the low- and middle-income countries face against the delta variant,” said Dr. Peter Hotez (https://www.texaschildrens.org/find-a-doctor/peter-jay-hotez-md-phd), Professor and Dean of the National School of Tropical Medicine at Baylor and Co-Director of the Texas Children’s Hospital Center for Vaccine Development.

“Widespread and global vaccination with our Texas Children’s-Baylor-BE vaccine would also forestall the emergence of new variants. We have previously missed that opportunity for the alpha and delta variant. Now is our chance to prevent a new global wave from what might follow.”

“Over the years, we have worked to make quality vaccines and pharmaceutical products accessible to families around the world. With this as our backdrop, we resolved to develop an affordable and effective COVID-19 vaccine. It has now become a reality,” said Mahima Datla, Managing Director, Biological E. Limited. “We deeply appreciate Texas Children’s Hospital Center for Vaccine Development, Baylor, CEPI, the Bill and Melinda Gates Foundation, and the government of India for their continuous support and cooperation during this journey. The combined efforts & unceasing support demonstrate that we can collectively overcome any challenge”.

Texas Children’s Hospital CVD received philanthropic support for the development of this vaccine from many generous donors across the country, including a significant contribution from The JPB Foundation in May 2020.

Also, "no-frills tech transfer"

The central government of India has already ordered 300 million doses. And BioE, the company manufacturing [Corbevax], plans to produce 100 million or more doses per month starting in February. Approximately 150 million doses have already been produced and are ready to roll out. In addition to what the company is supplying to India, BioE plans to deliver more than one billion additional doses to other countries.

What this means is that Corbevax will soon vaccinate more people than vaccine doses donated so far by the U.S. government or any other G7 country.

Corbevax is made using technology that has been employed worldwide for decades, meaning that manufacturing processes are generally already well-known and won’t require a steep learning curve like the one needed for the scale-up of new technologies such as mRNA, adenovirus and protein particle vaccines.https://www.scientificamerican.com/article/a-covid-vaccine-for-all/

Winehole23
12-31-2021, 10:55 AM
When compared with doses of the AstraZeneca–University of Oxford vaccine manufactured by the Serum Institute of India, CORBEVAX also produced a higher amount of neutralizing antibodies against the Delta and Beta variants of SARS-CoV-2, the virus that causes COVID (We expect to have Omicron data soon.) And it provided more durable and lasting protection. The vaccine neutralized variants of concern in laboratory animal studies (https://pubmed.ncbi.nlm.nih.gov/34268512/) and was highly protective in two nonhuman primate challenge trials. (https://www.science.org/doi/10.1126/sciimmunol.abh3634?url_ver=Z39.88-2003&rfr_id=ori:rid:crossref.org&rfr_dat=cr_pub%20%200pubmed)

As a recombinant protein vaccine developed from the receptor biding domain of the spike protein on the virus’s surface, combined with Dynavax Technologies’ CpG 1018 adjuvant with alum, the Texas Children’s CVD COVID vaccine can be stored using simple refrigeration. And like the hepatitis B vaccine, this COVID vaccine has one of the lowest costs of any available to date. No patents have been filed on the vaccine technology, and Texas Children’s CVD is assisting and co-developing the vaccine alongside BE and other vaccine producers in the Global South, which helps keep the cost low.

As a complement to vaccines, small-molecule therapeutic agents are needed to treat or prevent infections by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and its variants, which cause COVID-19. Affinity selection-mass spectrometry was used for the discovery of botanical ligands to the SARS-CoV-2 spike protein. Cannabinoid acids from hemp (Cannabis sativa) were found to be allosteric as well as orthosteric ligands with micromolar affinity for the spike protein. In follow-up virus neutralization assays, cannabigerolic acid and cannabidiolic acid prevented infection of human epithelial cells by a pseudovirus expressing the SARS-CoV-2 spike protein and prevented entry of live SARS-CoV-2 into cells. Importantly, cannabigerolic acid and cannabidiolic acid were equally effective against the SARS-CoV-2 alpha variant B.1.1.7 and the beta variant B.1.351. Orally bioavailable and with a long history of safe human use, these cannabinoids, isolated or in hemp extracts, have the potential to prevent as well as treat infection by SARS-CoV-2https://pubmed.ncbi.nlm.nih.gov/35007072/

Hard to understand why there hasn't been a warp speed effort for stuff like this, but similar is true for old tech, no frills vaccines like Corbevax.

It's almost as if protecting the profitability of 1st generation vaccine makers (i.e., presumably grateful/generous drug companies) trumped public health, but there's a simpler explanation. The all-important political requisite of "returning to normal" frames COVID mitigation as the problem to be overcome, rather than overcoming COVID itself. Converting a manageable public health issue into a background assumption of mass morbidity and death -- like the annual flu and highway deaths -- is the ostensible goal.

Bharat Biotech is among the world’s leading vaccine manufacturers. Its best known product, Covaxin, is authorized to prevent Covid in India and many other countries. But its experimental nasal vaccine may prove to be the real game changer.

Immunizing entire populations with a nasal or oral vaccine would be faster in the middle of a surge than injections, which require skill and time to administer. A nasal vaccine is likely to be more palatable to many (including children) than painful shots, and would circumvent shortages of needles, syringes and other materials.

Intranasal vaccines “can be administered easily in mass immunization campaigns and reduce transmission,” said Krishna Ella, chairman and managing director of Bharat Biotech.

There are at least a dozen other nasal vaccines in development worldwide (https://clinicaltrials.gov/ct2/show/NCT04816019), some of them now in Phase 3 (https://www.nytimes.com/interactive/2020/science/coronavirus-vaccine-tracker.html) trials. But Bharat Biotech’s may be the first to become available. In January, the company won approval (https://www.clinicaltrialsarena.com/news/bharat-biotech-intranasal-vaccine/) to begin a Phase 3 trial of the nasal spray in India as a booster for people who have already received two shots of a Covid vaccine.

Researchers at a South African biotechnology company say they have nearly created a copy of Moderna’s messenger-RNA-based vaccine against COVID-19, without Moderna’s involvement.

The company, Afrigen Biologics and Vaccines, in Cape Town, has made only microlitres of the vaccine, based on data that Moderna used to make its shot. But the achievement is a milestone for a major initiative launched by the World Health Organization (WHO) — a technology transfer hub meant to build capacity for vaccine manufacturing in low- and middle-income countries.

Our results suggest that CBD can block SARS-CoV-2 infection at early stages of infection, and CBD administration is associated with a lower risk of SARS-CoV-2 infection in humans. Furthermore, the active compound in patients is likely to be 7-OH-CBD, the same metabolite implicated in CBD treatment of epilepsy. The substantial reduction in SARS-CoV-2 infection risk of approximately an order of magnitude in patients who took FDA-approved CBD highlights the potential efficacy of this drug in combating SARS-CoV-2 infection. Finally, the ability of CBD to inhibit replication of MHV raises the possibility that CBD may have efficacy against new pathogenic viruses arising in the future.https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7987002/

Of course it was, it wasn't manufactured for Omicron, in any way, shape, matter, or form, but nobody said a fuckin' word about that when it came ashore as the New Year broke. 120k dead Americans later as February winds down the truth can finally emerge.

A new Covid vaccine shows 100 percent efficacy against severe disease and hospitalizations

Two doses of a new Covid vaccine that is based on a conventional approach achieved 100 percent efficacy against severe disease and hospitalizations, and

it could be an effective booster after other Covid shots,

The vaccine, made by the Europe-based pharmaceutical companies Sanofi and GSK,

The new vaccine had an efficacy of 75 percent against moderate-to-severe disease.

It showed 58 percent efficacy against symptomatic disease

in its Phase 3 clinical trial.

Although that number is lower than was observed for the mRNA vaccines made by Pfizer-BioNTech and Moderna in their initial trials, it is “in line with expected vaccine effectiveness in today’s environment dominated by variants of concern,”

"SaNOtize," a nitric oxide nasal spray

The spray is designed to kill SARS-CoV-2 in the upper airways. “It has proven anti-microbial properties with a direct virucidal effect on SARS-CoV-2. NONS, when sprayed over nasal mucosa, acts as a physical and chemical barrier against the virus, preventing it from incubating and spreading to the lungs,” said Glenmark.

Significantly, the spray is not “variant-specific”, and has been seen to work against the alpha, beta and delta variants of SARS-CoV-2, besides the Rhinovirus and H1N1 virus as well, said Tandon.

I remember telling Hater that variants weren't a problem because the beauty of the mRNA platform was how quickly the vaccine could be updated, 6-8 weeks they said. Here it's year 3 and they are pushing for a 4th dose of the wild original wild type vaccine. Boy did I get snookered :lol

It's hilarious how many dumbass leftists here call me a crackhead and whatnot but this "crackhead" must have the best immune system around tbh. Since May 2020 I haven't worn a mask, eat almost daily at restaurants, hangout with friends, have gone to many events, and come in contact with numerous people daily.

Not sure if this is what's helping me but I don't eat trash like the obese faction of our nation, I don't consume alcohol whatsoever, and I take plenty of vitamins daily. Maybe I'm just super lucky and my time will come soon but here's to more good times without that poisonous jab by bigGov & bigPharma. :tu

This pandemic was caused by humans dicking around with a virus. Nothing to discuss. Stop worshiping these people.

False. We already went through this, but we can revisit it again if you'd like.

"These people" saved countless lives, they deserve all the worshipping they get.
